Stem cells of the small and large intestine are marked by expression of the Wnt target gene LGR5, a leucine-rich-repeat-containing G protein-coupled receptor. Previous studies reported increased expression of LGR5 in human colorectal cancer (CRC) compared to normal tissue either by immunohistochemistry or in situ hybridization (ISH). However, as these studies were semi-quantitative they did not provide...
Single-cell measurements have broadened our understanding of heterogeneity in biology, yet have been limited to mostly observational studies of normal or globally perturbed systems. Typically, perturbations are utilized in an open-ended approach wherein an endpoint is assayed during or after the biological event of interest. Here we describe ShootingStar, a platform for perturbation analysis in vivo,...
The interaction between stationary localized states have long been studied, but localized states may undergo a number of instabilities that lead to more complicated dynamical regimes. In this case, the effects of the interaction are much less known. This chapter addresses the problem of the interaction between oscillatory and excitable localized states in a Kerr cavity. These oscillatory structures...
We find and characterize two different mechanisms that lead to excitability of localized structures in a Kerr cavity. In a first scenario the excitability appears a saddle loop bifurcation. In second place we show an excitability regime that arises trough a SNIC bifurcation.
