This study was conducted to investigate the influence of recipient cytoplasm on the development of somatic cell nuclear transfer (SCNT) embryos, using recipient oocytes and donor cells that were obtained from cats, cows, and pigs. Bovine and porcine oocytes were collected from ovaries obtained at a slaughterhouse, and cat oocytes were collected from ovaries obtained at local veterinary clinics following...
Investigations of the effect of selected preparations on in vitro growth of Trichoderma harzianum isolated from button mushroom (Agaricus bisporus) crop and causing green moulds, were conducted in 2007. Different concentration of the preparations were added to PDA medium. Control consisted of medium with no fungicides added. Diameter of the fungal colonies was measured after 2 and 4 days and at 6...
Experiments on somatic embryogenesis in selected spruce, fir and larch species were performed to determine if this method of micropropagation enables production of quality seedlings for forest nurseries. High frequencies of embryogenic callus in spruce (23-31%) and fir (29%) were achieved when mature zygotic embryos were used as explants, while in larches (36%) only when megagametophytes (endosperms...
The object of the research work was an isolate of S. hypericinum D 1224, as well as ten fungicides belonging to different chemical groups. The studies on the toxic effect were conducted in vitro by means of poisoning the media with the use of 1, 10 and 100 μg a.i./cm3 of the tested preparations. The obtained results showed that the compounds of thiophanatemethyl and triadimephon caused complete inhibition...
Placentae of Melandrium album excised from ovaries of unopened flowers were pollinated in vitro with pollen grains of Lychnis coronaria. Pollen grains germinated, and pollen tubes were found entering the micropyles 18 h after pollination. Double fertilization occurred. Immature and mature hybrid embryos at different stages of development were analyzed up to the third week of culture. Twenty-five days...
In order to estimate the best germination conditions of Encyclia aff. oncidioides seeds, five different media (Fast, Knudson C modified by Vajrabhaya, Murashige and Skoog, PB2 and modified Vacin and Went) with different concentrations of plant growth regulators such as benzyladenine (BA), naphthaleneacetic acid (NAA) and gibberellic acid (GA3) were tested. No beneficial effect was observed when BA...
Adventitious bud cultures were established by using buds of selected poplar clones (Populus tremula L. × P. alba L.) as initial explants. The Murashige and Skoog medium (1/2 and 1/4 MS) was used for multiplication and rooting of shoots. To the media, aluminium was added in the form of sulphate, at a concentration 50−70 mg Al dm-3. The culture was continued in vitro for more than 12 months. The cultures...
Adventitious bud cultures were established by using buds of selected poplar clones (Populus tremula L. × P. alba L.) as initial explants. The Murashige and Skoog medium (1⁄2 and 1⁄4 MS) was used for multiplication of shoots. Aluminium and copper were added to the medium in the form of sulphates. Copper, lead and aluminium were also added to the medium in the form of nitrates. Low concentrations of...
The effect of fructose, glucose, maltose and sucrose at various concentrations on Brassica napus embryos was studied. The morphogenetic reaction depended on the type and concentration of sugar in the medium. The frequency of developing embryos was highest on sucrose, followed by maltose and glucose. Fructose did not stimulate embryo growth. Spectrophotometry of autoclaved fructose showed an absorbance...
The peculiarities of seminal and vegetative multiplication in orchids are discussed. The main pathways of morphogenesis during the formation of the whole organism of orchids in natural conditions and in vitro culture are reviewed. Peculiarities of development and the role of the protocorm in the life cycle are pointed out. Special attention is paid to the phenomenon of polyembryony of orchids and...
Autonomous endosperm (AE) was found in 39 (17.3%) of in vitro cultured unpollinated ovaries of Brassica napus L. cv. Topas, with average frequency 1.5 ovule with AE per ovary. AE was induced on MS medium and on MS + 2 mg/l BAP + 0.1 mg/l NAA , MS + 2 mg/l BAP + 2 mg/l 2,4-D, and in ovules exposed to 1 h auxin treatment (MS + 20 or 40 mg/l 2,4-D) and then transferred to hormone-free MS medium. The...
Antagonistic effect of Bacillus polymyxa, strain S13, toward Fusarium oxysporum f. sp. tulipae was evaluated in vitro and in vivo. The growth of the pathogen was greatly inhibited in dual cultures with Bacillus polymyxa on potato dextrose agar. Suspension of B. polymyxa and its filtrate substantially inhibited spore germination and development of Fusarium oxysporum f. sp. tulipae on tulip bulbs.
