Contexto
Os receptores de canabinoide e o receptor – alfa ativado por proliferador de peroxissoma (peroxisome proliferator‐activated receptor‐alpha, PPAR‐α) têm sido reconhecidos como potenciais alvos terapêuticos para o tratamento de dermatopatias.
Hipótese/Objetivos
O objetivo deste estudo foi investigar a distribuição dos receptores de canabinoide tipo 1 e tipo 2 (CBR1 e CBR2) e PPAR‐α na pele felina e verificar a ocorrência de alterações no curso de dermatites por hipersensibilidade.
Animais
Doze gatos de proprietários. As amostras de pele foram coletadas de cinco gatos saudáveis sem lesões cutâneas e sete gatos diagnosticados clinicamente com dermatite por hipersensibilidade.
Métodos
As alterações histopatológicas foram investigadas nos cortes histológicos de pele corados com hematoxilina e eosina. Realizou‐se imunofluorescência indireta para CBR1, CBR2 e PPAR‐α nos blocos parafinados, e a especificidade dos anticorpos foi testada por análise de Western blot.
Resultados
Todas as amostras cutâneas de gatos com dermatite por hipersensibilidade receberam diagnóstico histopatológico de dermatite eosinofílica. Os receptores CBR1, CBR2 e PPAR‐α estavam presentes ao longo pele de ambos gatos normais e gatos alérgicos. Na pele de gatos normais, estes receptores estavam distribuídos principalmente no compartimento epitelial. Houve aumento na expressão dos receptores na hipersensibilidade, comparado à pele normal, e as principais alterações na distribuição foram suprabasal para CBR1, dérmica para CBR2 e expressão pronunciada de PPAR‐α na epiderme hiperplásica e infiltrado perivascular.
Conclusões e importância clínica
O aumento na expressão de receptores de canabinoide na pele de gatos com dermatite por hipersensibilidade sugere a existência de uma estratégia endógena protetora e pode corroborar com o uso de agonistas de receptores de canabinoide naturais ou de PPAR‐α no tratamento da dermatite felina por hipersensibilidade.
