In Zusammenarbeit der Fachgebiete Mikrobiologie und Hygiene, Milchhygiene, Fleischhygiene und Bakteriologie des BgVV wurden gemäß der derzeit vom BgVV empfohlenen Untersuchungsvorschrift für den Nachweis von VTEC in Milch 105 Rinderhackfleischproben, 72 Weichkäseproben aus Rohmilch und 47 Weichkäseproben aus pasteurisierter Milch aus dem Handel auf das Vorkommen von VTEC untersucht. Daneben wurde der allgemeine mikrobiologische Status der Proben festgestellt, um diesen mit den VTEC-Befunden in Beziehung setzen zu können. Zwanzig Prozent der untersuchten Rinderhackproben und 4,2 % der Rohmilchweichkäseproben wiesen in der PCR positive Reaktionen auf. Bei Anwendung eines kommerziellen ELISA-Tests verlief der Verotoxin (VT)-Nachweis in ebenfalls 4,2 % der Rohmilchkäseproben, jedoch nur in 14,3 % der Rinderhackproben positiv. Sämtliche Weichkäseproben, die aus pasteurisierter Milch hergestellt wurden, reagierten in der PCR und im ELISA-Test negativ. Das Vorkommen von verotoxinogenen E. coli war im Rinderhack mit hohen Gesamtkeimzahlen von>107 KbE/g und höheren Enterobacteriaceae-Keimgehalten (>105 KbE/g) sowie in den Rohmilchweichkäseproben mit hohen Coliformen-Keimgehalten assoziiert. Keine Beziehung bestand zwischen dem herkömmlichen Nachweis nicht-toxinogener E. coli (bei 44 °C) und dem Nachweis der VTEC. Aus 12,3 % der Rinderhack- und aus 2,8 % der Rohmilchweichkäseproben konnten Verotoxin-produzierende E. coli isoliert werden. Keiner der isolierten Stämme besaß alle Virulenzeigenschaften des klassischen verotoxinogenen E. coli O157. Es handelte sich mit einer Ausnahme nur um VT-2-Bildner. Bei weniger als 50 % der Stämme war das HLy-Gen zu detektieren, das eaeA-Gen konnte in keinem der isolierten Stämme nachgewiesen werden. Die Ergebnisse der Serotypisierung ausgewählter Stämme zeigen die starke Heterogenität der im Untersuchungsmaterial vorkommenden VTEC auf. Verotoxin-produzierende E. coli-Stämme, die in Deutschland bereits mit EHEC-Infektionen in Zusammenhang gebracht wurden (O157, O26, O22 und O111), konnten in dieser Studie aus den Rinderhack- und den Rohmilchweichkäseproben nicht isoliert werden.
