Frühere Studien haben die Effekte von Sclerostin-Antikörpern (Scl-Ab) und Parathormon (1–34, PTH) auf die Heilung von Osteoporose gezeigt, jedoch sind Berichte über die kombinierten Effekte von Scl-Ab plus PTH auf Osteoporose limitiert. Ziel dieser Studie war es, die Auswirkungen einer Kombinationsbehandlung mit Scl-Ab und PTH auf die Heilung einer Osteoporose bei ovarektomierten (OVX) Ratten zu untersuchen. Nach einer bilateralen Ovarektomie wurde zunächst 12 Wochen abgewartet, um die Standardbedingungen für Osteoporose im Tiermodell zu schaffen. Dann erhielten die Ratten randomisiert einen Träger (Kontrolle), Scl-Ab (25 mg/kg KG, 2‑mal wöchentlich), PTH (60 μg/kg KG, 3‑mal wöchentlich) oder PTH plus Scl-Ab bis zum Tod in Woche 12. Das Blut und die distalen Oberschenkelknochen der Ratten wurden zur Beurteilung entnommen. Die Ergebnisse der Osteoporosebehandlung wurden mittels einer Serumanalyse, Histologie, Mikro-Computertomographie (Mikro-CT) und biomechanischen Tests evaluiert. Die Ergebnisse dieser Studie zeigten, dass PTH + Scl-Ab stärkere Auswirkungen auf die Prävention und die Behandlung von Osteoporose bei OVX-Ratten haben als die jeweilige Monotherapie. Die Gruppe mit PTH + Scl-Ab zeigte die stärksten Effekte auf die Knochenvolumenfraktion (BV/TV), die Dicke der Knochentrabekel (Tb.Th), die Anzahl (Tb.N) und den Abstand (Tb.Sp) der Trabekel, die Knochenmineraldichte (BMD) sowie die Stärke des distalen Oberschenkelknochens und erhöhte Werte des Prokollagens Typ I N‑terminales Propeptid (PINP) und Osteokalzin. Im Gegensatz dazu zeigte die Monotherapie mit PTH oder Scl-Ab bei der Beurteilung der Metaphyse des kontralateralen Femur in der Histologie, im Serum, in biomechanischen Tests und im Mikro-CT keinen Unterschied zwischen den Behandlungsgruppen. Diese Ergebnisse deuten darauf hin, dass Scl-Ab plus PTH einen zusätzlichen Effekt auf die Osteoporose bei OVX-Ratten hat.
