Einleitung
Die Tatsache, dass die Drüsenzellen der Tränendrüse des Menschen PAS-positiv reagieren, ließ schon seit langer Zeit die Synthese von Mukosubstanzen vermuten. Erst kürzlich erfolgte jedoch eine erste, dem heutigen Kenntnisstand entsprechende Differenzierung.
Ergebnisse
Untersuchungen mit Hilfe der Polymerasekettenreaktion (RT-PCR) zeigten, dass die Muzine MUC1, MUC4, MUC5AC, MUC5B, MUC6 und MUC7 in der humanen Tränendrüse exprimiert werden; MUC2 wird dagegen in der Tränendrüse nicht synthetisiert. Immunhistochemische Studien konnten das membrangebundene Muzin MUC1 an der Zelloberfläche von Azinuszellen lokalisieren. Das sezernierte Muzin MUC5AC reagierte in Becherzellen des Ausführungsgangsystems der Tränendrüse posititiv. Die Muzine MUC5B und MUC7 ließen sich im Zytoplasma von Azinuszellen nachweisen und MUC7 darüber hinaus in den Epithelzellen der Ausführungsgänge. MUC2 (RT-PCR negativ) und MUC6 (RT-PCR teilweise positiv) konnten mittels immunhistochemischer Analysen nicht detektiert werden. Die mRNA-Expression des Muzins MUC4 war nur in 6 von 30 Tränendrüsenproben von Personen, die keine Tränenersatzmittel erhielten, nachweisbar. Im Gegensatz dazu zeigten alle Proben von Personen mit trockenem Auge spezifische Transkripte für MUC4. Immuno-Dot-Blot-Analysen demonstrierten einen deutlichen Anstieg der Färbeintensität von MUC4, MUC5AC und MUC5B in Tränendrüsen von Personen, die mit Tränenersatzmitteln behandelt wurden, im Vergleich mit den anderen untersuchten Proben.
Schlussfolgerung
Die Ergebnisse zeigen, dass die Tränendrüse ein ganzes Spektrum an Muzinen synthetisiert, von denen ein Teil eine Rolle in der Pathophysiologie des trockenen Auges zu spielen scheint.
