Fragestellung. Die Fragestellung der vorliegenden Untersuchung war, ob in vitro die Isolierung von auf unterschiedlichen Biomaterialien adhrenten osteoblastren Zellen mandibulren Ursprungs und eine Darstellung der Osteokalzin-Gen- und -Proteinexpression aus diesen isolierten adhrenten Zellen whrend der gesamten Kultivierungsdauer von 14 Tagen mglich ist. Material und Methoden. Die osteoblastren Zellen mandibulren Ursprungs wurden aus Knochenbiopsien von einem Patienten isoliert, vermehrt und auf drei unterschiedlichen Biomaterialien (Frios Algipore, PepGen P-15, OsteoGraf/LD-700) ohne Zusatz osteogener Faktoren kultiviert. Am 7. und 14. Tag wurden eine automatische Zellzahlmessung, eine Frbung der alkalischen Phosphatase und eine Bestimmung der Osteokalzin-Gen- und -Proteinexpression mittels RT-PCR und SDS-PAGE/Western Blotting durchgefhrt. Zustzlich erfolgte eine rasterelektronenmikroskopische Darstellung vor und nach der Isolierung der auf Biomaterialien adhrenten Zellen. Ergebnisse. Die Isolierung von auf Biomaterialien adhrenten osteoblastren Zellen mandibulren Ursprungs ist mittels der von uns angewandten Methode mglich. Auch die Bestimmung der Expression von Osteokalzin auf Gen- bzw. Proteinebene von auf Biomaterialien adhrenten osteoblastren Zellen war an beiden Untersuchungstagen erfolgreich. In der anfnglichen Inkubationsphase (7 Tage) zeigte die Expression von Osteokalzin keine strkere Abweichung zwischen den Biomaterialien. Danach (7. bis 14. Tag) kam es zu einer Vernderung des Expressionsmusters, wobei klare Unterschiede zwischen den einzelnen Biomaterialien erkennbar waren. Schlussfolgerung. Es konnte gezeigt werden, dass nach Isolierung von auf Biomaterialien adhrenten Zellen mandibulren Ursprungs eine ausreichende Menge an Untersuchungsmaterial zur Darstellung der Osteokalzin-Gen- bzw. -Proteinexpression vorhanden ist. Die Isolierung von ausschlielich auf Biomaterialien adhrenten Zellen dient als Grundlage fr weitere Untersuchungen des Wachstumsverhaltens osteoblastrer Zellen in vitro. Informationen ber das Gen- und Proteinexpressionsmuster dienen zur besseren Beurteilung der Eigenschaften der verschiedenen Biomaterialien.
A new approach to addressing difficult tissue reconstructive or replacement problems in the oral cavity is to engineer new tissue by using selective cell transplantation on polymer scaffolds. The current study characterized the osteoblastic nature of adherent mandibular cells on biomaterials, which could have a potential use as scaffolds for tissue engineering strategies. Cells of mandibular origin from one patient were cultivated on three different biomaterials (PepGen P-15, Frios Algipore, and OsteoGraf/LD-700) for 7 and 14days and osteocalcin expression was demonstrated by RT-PCR and SDS-PAGE/Western blotting. In order to explicitly characterize only the adherent cells on the biomaterials, we first separated the biomaterials with adherent cells from the culture plate before trypsinization. We could demonstrate that cell growth of adherent mandibular osteoblast-like cells was significantly higher on biomaterials with an organic component (PepGen P-15) in comparison to Frios Algipore and OsteoGraf/LD-700, respectively. In conclusion, only the explicit study of adherent cells at the gene and protein levels gives information about the osteoconductivity of biomaterials.
