The present study investigated the effects of dichlorodiphenyltrichloroethane (DDT), methoxychlor (MXC), and γ-hexachlorocyclohexane (γHCH, lindane) on gap junctional intercellular communication (GJIC) in cultured bovine oviductal cells. GJIC was evaluated by microinjecting fluorescent dye Lucifer Yellow and observing the inhibition of the spreading of dye into adjacent cells. After incubation for 1 h at 37°C, a dose-dependent inhibition of GJIC was observed over a concentration range of 16 to 128 μm DDT, MXC, or γHCH compared with nonexposed controls. A significant inhibition began at 32 μm DDT, MXC, or γHCH. After incubation for 5 h, a dose-dependent inhibition of GJIC was obtained in the concentration range from 8 to 64 μm of the pesticides. The first significant inhibitory effect on GJIC was caused by 8 μm DDT, 16 μm MXC, and 32 μm γHCH. The 128 μm concentration of the pesticides was toxic. At pesticide concentration of 64 μm, the decrease in dye-coupling observed was not due to lethal cell injury, as is indicated by the use of trypan blue dye exclusion. After removal of 64 μm DDT from the culture medium, intercellular communication was reestablished within 3 h. Measurement of cytosolic free Ca 2+ concentration [Ca 2+ ] i in fura-2/AM-loaded oviductal cells showed that the inhibition of GJIC by addition of DDT, MXC, or γHCH was not associated with a detectable increase in [Ca 2+ ] i . Coincubation of the DDT with dibutyryl-cAMP prevented the 64 μm DDT-induced inhibition of intercellular communication in adherent oviduct cells. It is suggested that organochlorine pesticides can influence cells responsible for reproduction.
Zusammenfassung: Hemmeffekte von Organochlorpestiziden auf den interzellulären Transfer von Lucifer Yellow bei kultivierten Eileiterzellen vom Rind. In vorliegender Studie wurden Effekte von Dichlordiphenyltrichlorethan (DDT), Methoxychlor (MXC) und γ-Hexachlorcyclohexan (γHCH, Lindan) auf die durch Gap junction vermittelte interzelluläre Kommunikation (GJIK) bei kultivierten Oviduktzellen vom Rind untersucht. Der Nachweis der GJIK erfolgte nach Mikroinjektion von Lucifer Yellow und wurde über den interzelluläre Transfer des Fluoreszenzfarbstoffes bestimmt. Eine dosisabhängige Hemmung der GJIK wurde nach einstündiger Inkubation bei 37°C durch DDT, MXC und γHCH im Konzentrationsbereich von 16 bis 128 μm und nach fünfstündiger Inkubation im Konzentrationsbereich von 8 bis 64 μm verursacht. Die ersten signifikanten Hemmeffekte auf die GJIK wurden nach einstündiger Inkubation durch 32 μm aller drei Noxen und nach fünfstündiger Inkubation durch 8 μm DDT, 16 μm MXC und 32 μm γHCH verursacht. Eine Konzentration von 128 μm aller Pestizide war nach einer fünfstündigen Inkubationszeit toxisch; während die signifikante Hemmung der GJIK bei 64 μm auf Grund der Trypanblaufärbung nicht auf lethale Zellschädigung zurückzuführen war. Darüberhinaus führte die Entfernung von 64 μm DDT aus dem Kulturmedium innerhalb von 3 h zur Wiederherstellung der GIJK. Die Messung der intrazellulären Ca 2+ -Konzentration [Ca 2+ ] i in fura-2/AM beladenen Oviduktzellen zeigte, daβ die Hemmung der GJIK nach Zugabe von DDT, MXC oder γHCH nicht durch eine erhöhte [Ca 2+ ] i verursacht wurde. Koinkubation von DDT mit dibutyryl-cAMP verhindert die 64 μm DDT-induzierte Hemmung der interzellulären Kommunikation bei adherenten Oviduktzellen. Aus den Ergebnisse kann geschluβfolgert werden, daβ Organochlorverbindungen Zellen, die für die Reproduktion verantwortlich sind, beeinflussen.
