The synthesis of some acyclic 1-[1,2,3-triazol-(4 and 5)-ylmethyl]-1H-pyrazolo[3,4-d]pyrimidine nucleosides, via 1,3-dipolar cycloaddition reaction, from 4-substituted N1-propargyl-1H-pyrazolo[3,4-d]pyrimidines 7a,b and azido derivative 8 is described. Compounds 7a,b and all deprotected acyclic nucleosides 11a,b, 12a,b and 13–22 were evaluated against: HIV-1, HIV-2 and a series of tumour cell-lines. No marked activity was found. Their anti-tuberculosis evaluation showed that compound 7b had marked activity.
Version française abrégée—Synthèse de quelques acyclonucléosides analogues de l'acyclovir. Les analogues des nucléosides naturels se sont révélés être de puissants agents antiviraux, grâce à leur capacité à inhiber l'ADN polymérase. La chimie de ces nucléosides est devenue un champ d'étude largement exploité dans la recherche de nouvelles drogues plus actives et moins cytotoxiques. Par conséquent, plusieurs familles d'analogues nucléosidiques ont été synthétisées et testées, tels que les acyclonucléosides [2, 3]. Parmi ces analogues, les acyclonucléosides pyrazolo[3,4-d]pyrimidiques restent, jusqu'à présent, un groupe peu étudié. À cet égard, nous avons reporté la synthèse d'une série d'acyclonucléosides pyrazolo[3,4-d]pyrimidiques [4] (figure 1). À la suite à ce travail, nous reportons, dans cette note, la synthèse des 1-[1-(2-hydroxyéthoxy)méthyl-1,2,3-triazol-(4 et 5)-ylméthyl]-1H-pyrazolo[3,4-d]pyrimidines substituées en position 4 (figure 2), afin de déterminer l'influence sur l'activité biologique du groupement 1,2,3-triazol-(4 ou 5)-ylméthyle, introduit entre les bases pyrazolo[3,4-d]pyrimidiques et le pseudo-sucre de l'acyclovir: le (2-hydroxyéthoxy)méthyle. En effet, les régio-isomères 9a,b et 10a,b, issus de la condensation des composés 7a,b, et le dérivé d'azoture 8, via la réaction de cycloaddition dipolaire [3+2], ont été séparés et déprotégés pour donner les acyclonucléosides 11a,b et 12a,b (schéma 2). Les produits 11a et 12a ainsi obtenus nous ont servi de précurseurs pour préparer les acyclonucléosides 13–22 (schéma 3). Les produits 7a,b, 11a,b, 12a,b et 13–22 ont été testés contre le VIH-1, le VIH-2 et contre une série des cellules tumorales. Aucune activité significative n'a été détectée. L'évaluation de l'efficacité de ces composés contre la tuberculose montre que le produit 7b présente une activité remarquable.
