Pyridoxal-5'-phosphate, a metabolic derivative of vitamin B6, was successfully used as a probe for rotational diffusion. As the intrinsic cofactor in glycogen phosphorylase b, in binding with bovine serum albumin and in mixed micelles, the Schiff base adduct exhibited transient absorption dichroism at μs times. Its usefulness in measuring the slow rotation of proteins and micelles was demonstrated.
