Using electrospray ionization mass spectrometry and combining charge state distribution observations with hydrogen/deuterium exchanges, it is possible to evidence the presence of several conformers or to obtain local information on conformation changes induced by single mutations on bacterial cytochromes c. Mass spectrometry is shown to be a fast and sensitive technique to get information on the three-dimensional structure of proteins.
