In our experiments a new anxiolytic from the group of 5-HT 3 antagonistsondansetron has been used in rats developing tolerance to oxazepam. After 7 days oxazepam administration a significant decrease of the anxiolytic behavior is observed in Crowley test. After additionally injected ondansetron (0.1 mg/kg i.p.) however the initial anxiolytic activity was restored or even surpassed. After 14 days oxazepam administration it's anxiolytic activity was diminished. A single injection of 0.1 mg/kg ondansetron restored the anxiolytic behavior (rise of BWT and WSE). After 21 days the anxiolytic activity of oxazepam was totally abolished and the single injection of ondansetron did not restore the state of anxiolysis. This shows that ondansetron was not able to overcome the anxiogenic effect accompanying the state of tolerance to oxazepam.
