In this report a low-cost illumination device for in vitro photodynamic treatment (PDT) is presented based on the use of light-emitting diodes mounted in an array-like arrangement. The diode array constructed in our laboratory is characterized in terms of irradiance as a function of the current applied, spatial homogeneity and effects of prolonged irradiation on media temperature. The efficiency for applications within in vitro PDT is demonstrated by analysis of the survival of the A431 human cancer cell line sensitized with either hypericin, meta-tetra (hydroxyphenyl) chlorin (mTHPC) or Photofrin and irradiated with appropriate diode types of the respective wavelength. According to the results obtained, a diode-based array for illumination represents an inexpensive technical alternative for stable, reproducible, homogeneous and specific irradiation for in vitro PDT experimentation.
In diesem Artikel wird eine kostengünstige Lichtquelle für photodynamische Behandlungen in vitro vorgestellt, welche aus einem Array aus Leuchtdioden (LED) besteht. Für dieses in unserem Labor hergestellte Array wurden die Leistungsdichte als Funktion der angelegten Stromstärke, die Homogenität des Beleuchtungsfeldes, sowie die Medienerwärmung nach längerer Bestrahlung bestimmt.Die Phototoxizität einer in vitro PDT unter Verwendung des LED Arrays wurde für die humane Karzinomzelllinie A431 bestimmt, die mit den drei Photosensibilisatoren Hyperizin, mTHPC (meta-tetra (hydroxyphenyl) chlorin) sowie Photofrin inkubiert und jeweils mit der spezifischen Anregungswellenlänge bestrahlt wurde.Unsere Resultate sprechen für die Verwendung von diodenbasierten Lichtquellen als kostengünstige, stabile, zuverlässige und spezifische Alternative zur homogenen Bestrahlung im Rahmen der in vitro PDT.
En este informe se describe un dispositivo de irradiación de bajo costo para la terapia fotodinámica (PDT) in vitro basado en el uso de diodos emisores de luz montados en forma de array. El arreglo de diodos construído en nuestro laboratorio, está caracterizado en términos de irradiancia como función de la corriente aplicada, la homogeneidad espacial y los efectos de la irradiación prolongada sobre la temperatura del medio. Para demostrar la eficiencia en diversas aplicaciones dentro de la PDT in vitro se analizó la sobrevida de células de la línea tumoral humana A431 sensibilizadas con Hipericin, mTHPC (metatetrahidroxifenilclorina) o Photofrin, e irradiadas con diferentes diodos a la longitud de onda apropiada. De acuerdo a los resultados obtenidos, este arreglo de diodos representa una alternativa técnica económica para una irradiación estable, reproducible, homogénea y específica en la experimentación con PDT in vitro.
