Comprendre le diabète et tenter de le guérir nécessite l’identification des régulateurs de la fonction beta pancréatique. Nous avons récemment démontré que la voie de signalisation des glucocorticoïdes inhibe le développement et la fonction des cellules beta, en faisant intervenir un co-régulateur transcriptionnel, PGC-1a. La surexpression de PGC-1a spécifiquement dans les cellules beta (souris Ins-PGC-1a) s’accompagne d’une intolérance au glucose associée à une diminution de la sécrétion d’insuline et de la masse de cellules beta. Puisque PGC-1a contrôle la biogénèse mitochondriale, notre objectif a été de définir comment la fonction mitochondriale et l’état énergétique des cellules beta pouvait participer aux défauts de masse et de fonction de ces cellules quand PGC-1a est surexprimé.Nous avons étudié, dans deux modèles de surexpression de PGC-1a (ilots de souris Ins-PGC-1a et lignée beta Min6), la sécrétion d’insuline en réponse au glucose, la fonction mitochondriale (respiration cellulaire, contenus en ATP, production d’espèces réactives de l’oxygène ou ERO) et l’état énergétique cellulaire par la phosphorylation de l’AMPK.Dans les cellules Min6 la surexpression de PGC-1a entraine un déficit d’insulino-sécrétion, une augmentation de la respiration cellulaire et de production d’ERO. La production d’ATP est normale, mais paradoxalement l’AMPK est activée. L’utilisation d’un antioxydant puissant, le Tempol, empêche la production d’ERO, l’activation de l’AMPK et restaure une insulino- sécrétion normale. De plus, l’inhibition de l’AMPK par le composé C permet également de restaurer une insulino-sécrétion normale. Les ilots des souris Ins-PGC-1a présentent également un défaut d’insulinosécretion associé à une production accrue d’ERO et une activation d’AMPK et l’utilisation d’un antioxydant permet de restaurer partiellement l’insulino-sécrétion.La surexpression de PGC-1a entraîne un stress oxydant capable d’activer l’AMPK et cette cascade d’événements inhibe la sécrétion d’insuline, démontrant le lien fort entre stress oxydant et stress énergétique dans la cellule beta.
