Un modele de bacteriemie experimentale animale est propose. Le microporc Yucatan, outre ses caracteristiques cliniques, biologiques et pharmacocinetiques proches de l'homme, est un animal docile et de grande taille autorisant des prelevements sanguins iteratifs et de volumes importants. Six microporcs femelles adultes sont inocules par voie intraveineuse avec une suspension d'Escherichia coli contenant en moyenne 5,1 x 10 9 bacteries. L'evolution est spontanement suivie durant cinq jours, tant sur le plan bacteriologique que clinique et biologique. Un syndrome inflammatoire systemique, un sepsis, une insuffisance d'organe et des hemocultures positives au meme germe que celui qui a ete inocule apparaissent des les premieres heures qui suivent l'inoculation, mimant la maladie humaine. Une apathie, une incoordination motrice, une anorexie, une tachypnee, un etat febrile a plus de 38 o C, une leuconeutropenie suivie d'une polynucleose neutrophile, une anemie, une thrombopenie, une tubulonephrite aigue et une acceleration de la vitesse de sedimentation est observee chez tous les animaux. Certains presentent en outre une elevation de la proteine C reactive, d'autres une augmentation de l'IL-6 et de l'IL-8. Au terme de l'etude tous les animaux sont vivants et les hemocultures de cinq microporcs sont toujours positives. Ce modele chronique, reproductible est propose pour l'evaluation des traitements antibacteriens.
The Yucatan micropig has been used to develop an experimental model of chronic bacteremia. This animal exhibits clinical and biological characteristics that are close to those in humans, and the pharmacokinetic behaviours of many classes of drugs in this model are similar to those in man. Six adult females were intravenously inoculated with a mean Escherichia coli inoculum of 5.1x10 9 bacteria. During five days of spontaneous evolution, the medical follow-up includes biological, clinical and bacteriological parameters. A systemic inflammatory syndrome, a sepsis, an organ insufficiency and positive blood cultures mimic the human disease. In all animals there is an adynamia, a lack of motor coordination, an anorexia, a tachypnea, a fever, a leuconeutropenia followed by an hyperleucocytosis, an anemia, a thrombopenia, an acute tubulonephritis and an elevated sedimentation rate. In some cases, there is an increase of the C reactive protein, in others, an increase of IL-6 and IL-8. At day five, all animals are alive, and five micropigs have positive blood cultures. This chronic, reproducible model is thus suitable for further antibacterial treatments evaluations.
