We have established experimental models of bovine leukemia virus (BLV) infection followed by progression to persistent lymphocytosis (PL) positive (BLV + PL + ) or PL negative (BLV + PL − ) stages of infection. Two out of six BLV infected animals developed PL + 4 weeks after BLV infection. One other animal became PL + late in the course of infection and three infected animals stayed PL − . These animals (PL − ) exhibited transient lymphocytosis 3–4 weeks after infection and sustained PL − lymphocyte counts up to 24 weeks after infection. Competitive RT-PCR analysis of IFN-γ mRNA expression revealed that peripheral blood mononuclear cells (PBMC) of animals with PL + status developed by 4 weeks after infection had augmented IFN-γ mRNA expression 3–4 weeks after BLV infection. However PBMC of animals that sustained a long-termed PL − lymphocyte count had elevated IFN-γ mRNA expression 1–24 weeks after infection. Competitive RT-PCR analysis of IL-2 mRNA expression showed an increase in the levels of IL-2 mRNA in PL − animals. Interleukin-10 (IL-10) mRNAs expression were elevated both in PL + and PL − animals from 3 and 12 weeks after infection respectively. We suggest that early and extended expression of cellular response cytokines may delay the progression to PL + in enzootic bovine leukemia.
Nous avons établi un modèle d’infection experimentale avec le virus de leucémie bovine (BLV) conduisant à une lymphocytose permanente (BLV + PL + ) ou lymphocytose absente (BLV + PL − ). Deux des 6 veaux infectés avec BLV ont développé l’état de PL + à partir de 4 semaines après l’infection. Un autre animal devint PL + longtemps après l’infection et 3 autres sont restés PL − . Ces 3 animaux (PL − ) montrèrent une lymphocytose transitoire 3–4 semaines suivant l’infection et maintinrent l’état PL − jusqu’à 24 semaines aprés. L’analyze par RT-PCR de l’expression de INF-γ m RNA a relevé que les cellules mononucléaires du sang périphérique (PBMC) des animaux qui ont démontré l’état PL + 4 semaines après l’infection avaient une augmentation d’expression de l’INF-γ mRNA, 3–4 semaines après l’infection. Les PBMC des animaux montrant une lymphocytose soutenue (PL − ) montraient une expression élevée de l’INF-γ mRNA à partir d’une à 24 semaines suivant l’infection. Par analyse compétitive du RT-PCR pour l’expression d’IL-2mRNA, on a remarqué une augmentation des niveaux d’IL-2 mRNA chez les PL − animaux. L’expression de l’interleukine-10 (IL-10) était élevée chez les veaux PL + de même que chez ceux avec PL − de 3 à 12 semaines respectivement. Nous suggérons retarder le progrés de PL + dans le cas de la leucémie bovine énzootique.
