Les canaux K ATP , constitués des sous-unités Kir6.2 et SUR1, sont considérés comme la cible principale du glucose pour le contrôle de la sécrétion d’insuline (SI). Leur fermeture provoque une dépolarisation et aboutit à une élévation de la concentration cytosolique de Ca 2+ ([Ca 2+ ] c ) qui déclenche la SI. Il a cependant été suggéré récemment que le glucose pourrait moduler la [Ca 2+ ] c et la SI en l’absence de canaux K ATP (souris SUR1 KO). Dans cette étude nous avons caractérisé les effets du glucose dans des îlots de souris Kir6.2 KO.La SI et la [Ca 2+ ] c ont été mesurées lors de perifusions des îlots (fraîchement isolés ou cultivés pendant 18 h) avec 1 ou 15 mM glucose (G1-G15).En présence de G1, la [Ca 2+ ] c et la SI d’îlots Kir6.2 KO frais sont modérément augmentées par rapport à des îlots témoins (avec canaux K ATP ). Une stimulation par G15 augmente la [Ca 2+ ] c et la SI de façon modeste mais étonnamment biphasique. La culture des îlots en G5 accentue ces changements biphasiques de la [Ca 2+ ] c et de la SI provoqués par une stimulation aiguë avec G15. Après culture en G10, la [Ca 2+ ] c et la SI sont déjà augmentées en présence de G1. L’augmentation biphasique de la [Ca 2+ ] c et de la SI provoquée par le glucose dans des îlots Kir6.2 KOest due à une stimulation de l’influx de Ca 2+ par des canaux voltage-dépendants de type L. La [Ca 2+ ] c des cellules β Kir6.2 KOprésente des oscillations semblables à celles observées en présence de canaux K ATP et modulées par le glucose.L’inactivation séparée de Kir6.2 et de SUR1 induit un même phénotype des cellules β. Contrairement aux modèles couramment acceptés, nos résultats suggèrent que les canaux K ATP ne sont pas les seuls transducteurs des changements du métabolisme en changements du potentiel de membrane, de la [Ca 2+ ] c et de la SI. Une régulation K ATP -indépendante du signal déclenchant la SI est possible de façon aiguë et chronique.
