This project focused on the effects of aflatoxin B1 (AFB1), a food-contaminating mycotoxin produced by fungi, genus Aspergillus, on the release and genetic expression of some important cytokines, i.e., (interleukin-1α (IL-1α), IL-6, tumor necrosis factor-α (TNFα)) by human monocytes. Monocytes, preincubated for different time periods with concentrations of AFB1 ranging from 0.01 to 1.0<space>pg/mL, were then activated with bacterial lipopolysaccharide. Cytokine levels were measured by immunoassay and mRNA by cDNA amplification. Pretreatment of monocytes with AFB1 resulted in a decrease in IL-1, IL-6 and TNFα release already at a concentration of 0.05<space>pg/mL. The gene expression of the cytokines considered was drastically affected by treatment with AFB1. In fact, AFB1 completely blocked the transcription of IL-1α, IL-6 and TNFα mRNAs, while it did not affect β-actin mRNA at the concentrations used. It therefore appears that AFB1 exerts its effect on cytokine release through selective inhibition of specific mRNA, without affecting general protein synthesis.
Quelques metabolites secondaires du genre Aspergillus exercent des effets biologiques sur les monocytes humains. Nous avons etudie les effets de l'aflatoxine B1 (AFB1) (une mycotoxine produite par Aspergillus, moississure des aliments) sur le relachement et l'expression genetique de quelques cytokines importantes (IL-1α, IL-6, TNFα) produites par les monocytes humains. Les monocytes, pretraites par l'AFB1 a des concentrations s'echelonnant de 0,01 a 1,0<space>pg/mL, ont ete actives, par la suite, par le lipopolyoside bacterien (LPS). Le niveau des cytokines a ete evalue par des techniques immunoenzymatiques, et les ARNm, par amplification des ADNc. Le pretraitement des monocytes par l'AFB1 a provoque une diminution du relachement d'IL-1α, d'IL-6 et de TNFαdont les doses de depart sont aussi faibles que 0,05<space>pg/mL. L'expression genetique des cytokines etudiees a ete drastiquement frappe par le traitement par l'AFB1. En effet, l'AFB1 bloque completement la transcription des ARNm des cytokines etudiees, tandis qu'elle n'exerce aucun effet sur l'ARNm de la β-actine, aux concentrations utilisees. Ces resultats semblent demontrer que l'action de l'AFB1 sur le relachement des cytokines s'exerce par une inhibition selective des ARNm specifiques, sans agir sur la syntese proteique generale.
