Square wave voltammetry has been applied to the studies of the interactions between dopamine and DNA in order to estimate the binding constant and the binding site size using the Scatchard binding model. The concentrations of bound and free dopamine corresponding to the limits of slow and fast binding kinetics have been determined. Subsequently, the following binding parameters were found for the static and dynamic binding equilibria, respectively: K = 1.65 (š 0.4) × 105 L mol-1, s = 22.7 (š 1.9) and K = 5.32 (š 0.7) × 105 L mol-1, s = 17.1 (š 1.1) (K - binding constant, s - effective number of base pair involved in binding). The influence of the ionic strength revealed on the equilibria is indicative for electrostatic attraction as the main binding force between dopamine and DNA. Some contributions from hydrogen bonding and intercalation have been also detected.
Badano odziaiywania między dopaminą i DNA za pomocą woltamperometrii fali prostokątnej. Celem badań było oszacowanie stałej wiązania i rozmiaru miejsca wiążącego na DNA. Wykorzystano model Scatcharda. Wyznaczono stężenie wolnej (związanej dopaminy odpowiadające granicom wolnej i szybkiej kinetyki wiązania. Następujące parametry wiązania obliczono dla odpowiednio - statycznej i dynamicznej równowagi wiązania: K = 1.65(š 0.4) x 10(5) L mol(-1), s = 22.7(š 1.9) i K = 5.32(š 0.7) x 10(5) L mol(-1), s = I7.1( š 1.1) gdzie K to stała wiązania, a s to efektywna liczba zaangażowanych par zasad. Wpływ mocy jonowej wskazuje, że główną silą wiążącą są oddziaływania elektrostatyczne jednakże powstawanie wiązań wodorowych interkaiacyjnych również wykryto.
