W ostatnich latach zaobserwowano znaczny wzrost liczby epidemii chorób pokarmowych związanych ze spożywaniem świeżych produktów, w skład których wchodzą surowce roślinne. Źródło zakażeń najczęściej wiązano z pomidorami, melonami, sałatą, pistacjami, przyprawami, kiełkami, malinami, roślinami strączkowymi i zbożem, a główne patogeny będące ich przyczyną to: Salmonellaspp.,patogenne Escherichia coli, Campylobacterspp., Shigellaspp., Yersinia enterocolitica i Yersinia pseudotuberculosis, Listeria monocytogenes, Staphylococcus aureus, Bacillus cereus, Clostridium botulinum i Clostridium perfringens, a także norowirusy i HAV. Obecnie najbardziej obiecujące metody wykrywania i identyfikacji bakterii zanieczyszczających żywność opierają się na technice PCR. Metody wykorzystujące technikę real-time PCR (qPCR) pozwalają zdecydowanie szybciej niż metody klasyczne wykryć obecność określonego drobnoustroju w badanej próbce. Opracowanie niezawodnych i powtarzalnych metod wykrywania drobnoustrojów potencjalnie patogennych dla człowieka, opartych na technice qPCR, wymaga optymalizacji i standaryzacji oraz potwierdzenia kompetencji w badaniach biegłości. Zapotrzebowanie na szybkie i mniej pracochłonne metody wykrywania drobnoustrojów patogennych w żywności ciągle rośnie, co wpływa na rosnącą popularność metod opartych na qPCR.
Recently there has seen a significant increase in the number of outbreaks associated with fresh plant products. Source of outbreaks most often has been associated with the following crops: tomatoes, melons, lettuce, pistachios, spices, sprouts, raspberries, pulses and grain, and the main pathogens which cause them are: Salmonella spp., pathogenic Escherichia coli, Campylobacter spp., Shigella spp. Yersinia enterocolitica and Yersinia pseudotuberculosis, Listeria monocytogenes, Staphylococcus aureus, Bacillus cereus, Clostridium botulinumand Costridium perfringens, noroviruses and HAV. Currently, the most promising methods for detection and identification of food pathogenes are based on PCR. Methods based on the technique of real time PCR (qPCR) allow you to specify the presence of the organism in the sample, much faster than classical methods. The development of reliable and reproducible methods for the detection of potentially pathogenic microorganisms to humans based on qPCR technique requires optimization, standardization, and accurate assessment of proficiency testing. The demand for rapid, less labor-intensive and explicit methods for detecting specific microorganisms in foods is still growing which affects the growing popularity of qPCR-based methods.
