We have isolated agosterol A (1) from a marine sponge of Spongia sp. as a reversing substance to multidrug resistance (MDR) in human carcinoma cell lines, KB-C2 and KB-CV60, overexpressing P-glycoprotein and MRP, respectively. Agosterol A (1) has been isolated from a marine sponge of Spongia sp. and the absolute stereostructure elucidated. Agosterol A (1) is a novel polyhydroxylated sterol acetate, which completely reverses multidrug resistance in human carcinoma cells caused by overexpression of two kinds of membrane glycoprotein.
