Les cytokines de la famille IL-1 jouent un rôle très important dans l’immunité et l’inflammation, et constituent par conséquent des cibles de choix pour les maladies inflammatoires chroniques. Ainsi, des inhibiteurs de l’IL-1 sont actuellement utilisés en clinique pour le traitement de la polyarthrite rhumatoïde. L’IL-33, la dernière née des interleukines, présente des homologies structurales avec l’IL-1 et se lie au récepteur ST2 (IL-1R4), un récepteur de la famille IL-1 exprimé par les cellules inflammatoires impliquées dans l’allergie (lymphocytes Th2, mastocytes, basophiles, éosinophiles, cellules iNKT). De par sa capacité à déclencher des réponses inflammatoires de type allergique, l’IL-33 pourrait être un acteur majeur dans l’asthme et les autres maladies allergiques (dermatite atopique, rhinite allergique…).

IL-33, the most recent addition to the IL-1 family, has structural homologies with IL-1 and binds to the ST2 receptor (IL-1R4), a member of the IL-1 receptor superfamily expressed by inflammatory cells involved in allergy (Th2 lymphocytes, mast cells, basophils, eosinophils, iNKT cells). Because of its capacity to induce allergic-type inflammatory responses, IL-33 could be a major factor in asthma and other allergic diseases (atopic dermatitis, allergic rhinitis…).
