L’œuf est responsable chez l’enfant d’allergies parfois sévères liées à la présence d’IgE dirigées contre les 5 principaux allergènes de l’œuf. La protéine OVAX, homologue à l’ovalbumine, a été récemment identifiée comme possédant des propriétés antimicrobiennes mais la capacité de cette molécule à induire, déclencher ou même réguler une allergie doit être étudiée avant son utilisation comme conservateur dans l’industrie agroalimentaire. L’objectif de notre travail est d’étudier l’immunogénicité de l’OVAX et d’obtenir des anticorps monoclonaux humains spécifiques des différentes protéines de l’œuf.À partir de sérum de patients allergiques à l’œuf, nous avons évalué par Elisa l’immunogénicité de l’OVAX, en comparaison avec l’allergénicité de l’OVA. Afin de produire des anticorps monoclonaux humains, des lymphocytes B issus de patients allergiques au blanc d’œuf ont été immortalisés et clonés. La monoclonalité a été contrôlée par PCR, la spécificité et la caractérisation de cet anticorps ont été étudiées par Elisa et western blot.Nous avons mis en évidence, chez certains patients, la présence à la fois d’IgG et d’IgE dirigées contre l’OVAX et l’OVA suggérant ainsi une allergénicité croisée. Nous avons isolé un clone d’intérêt (clone 6H4) qui reconnaît à la fois l’OVA et l’OVAX suggérant l’existence d’un épitope commun entre ces deux protéines. Nous avons confirmé que notre clone 6H4 ne se fixe pas à l’ovomucoïde. Enfin, l’anticorps reconnaît un épitope séquentiel et a une affinité plus importante vis-à-vis de l’OVAX par rapport à l’OVA.Ces résultats montrent que les patients allergiques à l’œuf peuvent présenter des IgG mais surtout des IgE spécifiques de l’OVAX. Ces résultats s’expliquent en partie par la réactivité croisée des anticorps pour l’OVA et l’OVAX. Par ailleurs, nous avons pu produire un anticorps monoclonal humain dirigé contre l’OVAX, qui devrait nous permettre d’étudier, dans un second temps, l’allergénicité de l’OVAX et de développer des outils de diagnostic en santé humaine sans dépendre des sérums de patients. Ces outils permettront également le développement de tests diagnostiques pour les industriels concernés par les ovoproduits dont l’OVAX.
